Test Principle

ELISA 키트는 샌드위치-ELISA 원리를 사용합니다. 이 키트에 포함된 마이크로 ELISA 플레이트는 쥐 IL-10에 특이적인 항체로 사전 코팅되어 있습니다. 표준물질 또는 샘플을 마이크로 ELISA 플레이트 웰에 추가하고 특정 항체와 결합합니다. 그런 다음 쥐 IL-10에 특이적인 비오티닐화 검출 항체와 아비딘-홀세라디시 퍼옥시다제(HRP) 접합체를 각 마이크로 플레이트 웰에 연속적으로 첨가하고 배양합니다. 유리 성분은 씻어냅니다. 기질 용액을 각 웰에 추가합니다. 쥐 IL-10, 비오티닐화 검출 항체 및 Avidin-HRP 접합체가 포함된 웰만 파란색으로 표시됩니다. 효소-기질 반응은 정지 용액을 첨가하여 종료되고 색이 노란색으로 바뀝니다. 광학 밀도(OD)는 450nm ± 2nm의 파장에서 분광광도계로 측정합니다. OD 값은 Rat IL-10의 농도에 비례합니다. 시료의 OD를 표준 곡선과 비교하여 시료 내 쥐 IL-10의 농도를 계산할 수 있습니다.
 

Storage

An unopened kit can be stored at 2-8℃ for 1 month. If the kit is not supposed to be used within 1 month, store the components separately according to the following conditions once the kit is received.
 

Precision

Intra-assay Precision (Precision within an assay): 낮은, 중간 범위, 높은 수준의 쥐 IL-10을 가진 3개의 샘플을 각각 한 플레이트에서 20회 테스트했습니다.
Inter-assay Precision (Precision between assays): 낮은, 중간 범위 및 높은 수준의 쥐 IL-10을 가진 3개의 샘플을 3개의 다른 플레이트에서 각 플레이트에 20개의 복제본으로 테스트했습니다.
